Abstract
Nosema ceranae is a microsporidian parasite of the honey bee, Apis mellifera, that is widespread and causes extensive loss. A micro PCR-based detection method, termed ultra-rapid real-time PCR, was developed to rapidly and quantitatively detect Nosema ceranae in honeybees. A specific detection primer set(Nosema-180-F/R) was used to amplify a unique 180bp DNA fragment with a micro-scale chip-based real-time PCR system,(GenSpector 䠶 TMC-1000), which has uncommonly fast heating and cooling rates(10° C/second) and a small reaction volume(6µl). Real-time PCR assays using serially diluted DNA templates showed a high sensitivity of 102copies. As a result, the minimum detection time was 12 min and 19 sec(30 cycles), including a melting temperature analysis. Therefore, this novel detection method could be used as a rapid real-time PCR-based diagnostic tool and is expected to be applied to rapidly detect various pathogens.
