Abstract
Deformed wing virus (DWV) is associated with wing deformity of honeybee and is transmitted through Varroa destructor during the pupal stages. For easy detection of DWV, a DWV-specific loop-mediated isothermal amplification detection method (DWV-LAMP) was developed. DWV-LAMP employs Bst DNA polymerase and a set of four specially designed primers named DWV_LAMP_ F3/B3/FIP/BIP,which recognize a total of six distinct sequences in a DWV-specific sequence. Expensive equipment such as a thermo-cycler is not necessary to acquire a high level of precision. In our results, DWV cDNA was successfully amplified under isothermal conditions at about 54°C within 30 minutes. DWV-LAMP not only is expected to be used for DWV field detection but also can be applied to the development of field detection methods for various viral pathogens of animals.
