Abstract
Sacbrood virus (SBV) is one of most important pathogenic virus against honeybee. SBV-infection in honeybee could be mainly detected by PCR based method. Due to retro-viral life cycling, base exchanges of SBV genome seems to be best reason to fail in the PCR detection. In this study,structural gene Capsid protein region on the SBVgenome were cloned and determined genome sequence from 3 geographically different areas in Korea, and new SBV-specific primers were designed, named F/R296-SBV, based on genomic sequence from 3 Korean isolates. After optimization of PCR conditions, SBV-specific PCR using F/R296-SBV shows high sensitivity and accuracy in the test using SBV-infected sample and even multiply infected field samples. SBV-specific PCR using F/R296-SBV would be good alternative tool for the detection of SBV from honeybee.
