Abstract
Royal Jelly produced by Apis mellifera has been accepted and broadly used as a health promoting substance. Major Royal Jelly Protein 1 (MRJP1) is the most abundant protein that comprises 48% of the water soluble protein in Royal Jelly. The MRJP1 gene originated from Apis mellifera was cloned from its cDNA, and was expressed in E. coli by using pET-system. The purified recombinant-MRJP1 (rMRJP1) was used as antigen to generate specific monoclonal antibody. After cell-fusion and screening of hybridomas by indirect ELISA, the cloning was performed by limit dilution. Finally, two individual hybridomas produced specific monoclonal antibody (mAb) against rMRJP1and natural MRJP1. Using these clones, ascites fluids were generated separately in mouse. The fluids can be used to detect rMRJP1 antigen by ELISA and Western Blot at dilution rate 1/5000 without any unspecificity. These mAb would be useful not only for the detection and quantification of MRJP1 in Royal Jelly, but also for the qualification test of Royal Jelly itself.
